What is Glucose-6-Phosphate?

Glucose-6-phosphate (G6P) is a phosphorylated form of glucose and the first intermediate produced when glucose enters a cell. Hexokinase (or glucokinase in the liver) adds a phosphate group to glucose, trapping it inside the cell and committing it to metabolism.

G6P sits at a critical metabolic crossroads. From this single molecule, glucose can be directed into energy production (glycolysis), antioxidant defense (pentose phosphate pathway), energy storage (glycogen synthesis), fat production (de novo lipogenesis), or protein modification (hexosamine pathway).

G6P is primarily an intracellular metabolite and is not part of routine clinical blood panels. It is measured through specialized metabolomics testing, mainly in research settings.

Where Does Glucose-6-Phosphate Come From?

G6P is produced inside cells, primarily in the liver and muscle, when hexokinase or glucokinase phosphorylates incoming glucose. It is also generated during glycogen breakdown (glycogenolysis) and gluconeogenesis.
