What is Interleukin-4?
Interleukin-4 (IL-4) is a signaling protein (cytokine) that helps regulate the immune system. It is the key driver of the T-helper type 2 (Th2) immune response, which is the branch of immunity involved in allergic reactions and defense against parasites.
IL-4 acts on a wide range of cell types, including T cells, B cells, macrophages, and epithelial cells. It circulates at very low concentrations in healthy individuals, often near or below the detection limit of standard assays.
Measuring IL-4 in the blood can help assess Th2 immune activation and is commonly included in multiplex cytokine panels alongside related markers such as IL-5, IL-6, IL-10, IL-13, and TNF-alpha.
Where Does Interleukin-4 Come From?
IL-4 is produced primarily by Th2 helper T cells, with significant contributions from basophils, mast cells, eosinophils, natural killer T (NKT) cells, and type 2 innate lymphoid cells (ILC2s).
