What is Transforming Growth Factor Beta 1?
Transforming Growth Factor Beta 1 (TGF-β1) is a multifunctional cytokine that regulates immune responses, wound healing, cell growth, and tissue remodeling. It is the most abundant of the three TGF-β isoforms and belongs to a large superfamily that includes activins, inhibins, and bone morphogenetic proteins.
TGF-β1 circulates predominantly in a latent form bound to carrier proteins. Less than 1% exists as the free, biologically active form at any given time. Activation requires enzymatic cleavage or other triggers such as reactive oxygen species, integrins, or pH changes.
Platelets store extremely high concentrations of TGF-β1 in their alpha granules. Because clotting activates platelets and releases this stored TGF-β1, serum values are roughly 10 times higher than plasma values. Clinical testing uses K2EDTA plasma to measure true circulating levels.
Where Does Transforming Growth Factor Beta 1 Come From?
Nearly all cell types produce TGF-β1, but platelets are the largest reservoir, storing it at concentrations 40 to 100 times higher than other tissues. Macrophages, lymphocytes, fibroblasts, epithelial cells, and endothelial cells are also significant sources.
