What is Coproporphyrin I?
Coproporphyrin I is a byproduct of heme synthesis, the metabolic pathway that produces the heme component of hemoglobin and other essential proteins. It belongs to a family of compounds called porphyrins, which are intermediate molecules in this pathway.
Under normal conditions, the enzyme uroporphyrinogen III synthase directs heme production along the correct pathway. When this enzyme is insufficient or the pathway is disrupted, hydroxymethylbilane undergoes spontaneous cyclization to form uroporphyrinogen I, which is then converted to coproporphyrinogen I. This type I isomer is a metabolic dead-end and cannot be used to make heme.
Small amounts of coproporphyrin I are produced normally and excreted in urine and bile. It is measured alongside other porphyrins (including coproporphyrin III) as part of a quantitative urine porphyrin panel. The ratio of coproporphyrin I to coproporphyrin III is often more diagnostically informative than the absolute level alone.
Where Does Coproporphyrin I Come From?
Coproporphyrin I is produced primarily in the bone marrow and liver as a byproduct of heme biosynthesis. It forms when the intermediate hydroxymethylbilane undergoes spontaneous (non-enzymatic) cyclization instead of being directed along the normal pathway by uroporphyrinogen III synthase.
