What is Deoxyuridine Monophosphate?
Deoxyuridine monophosphate (dUMP) is a pyrimidine nucleotide that serves as the direct building block for making thymidylate (dTMP), one of the four essential components of DNA. The enzyme thymidylate synthase converts dUMP into dTMP using a folate-derived cofactor called 5,10-methylenetetrahydrofolate.
This conversion is the sole de novo pathway for producing thymine nucleotides in human cells, making dUMP a critical checkpoint in DNA synthesis. When this pathway is impaired, dUMP accumulates and uracil is mistakenly incorporated into DNA instead of thymine, leading to DNA strand breaks and impaired cell division.
Measuring dUMP (or its dephosphorylated form, deoxyuridine) provides a functional window into whether the folate and vitamin B12 pathways that drive DNA synthesis are working properly.
Where Does Deoxyuridine Monophosphate Come From?
dUMP is produced inside all dividing cells through two main routes: the reduction of uridine diphosphate (UDP) by ribonucleotide reductase, and the deamination of deoxycytidine monophosphate (dCMP) by dCMP deaminase. It is also generated when the protective enzyme dUTPase breaks down dUTP to prevent uracil from being built into DNA.
